Not necessarily - Jacoulet was notoriously unorthodox in numbering his prints, and his system of numbering was bizarre. There are certainly proof impressions but they are not typically encountered - unless one argues that every unnumbered Jacoulet impression is a proof, which I doubt very much. I have seen too many unnumbered specimens for that to be likely. I would argue that based on current research, some regular edition impressions do not have number stamps, which, but the way, would appear on the back of the print. So if your print is numbered, you would see the number stamp on the back, and thus you'd have to remove the prints from the frames (probably a good idea anyway as you should check on the condition of the prints, back and front (e.g., is the matting acid-free? are the prints pasted down? is there any mold or staining?). Please do not assume that the original frames and mats are archival or safe for your prints.
